Question for Photoshop wizards

A Question for Photoshop wizards:

Can you replace a color with "transparent"? I don't mean erasing an area of a layer. I mean selecting a color and replacing it with "transparent".

if you have the psd , transparent is just transparent you can add a new layer as the first layer and change background color

Technically, making a pixel transparent would be the same as deleting it.

You probably are looking for a mask. What I would do is use the select color range to get your selection, invert it (ctrl-shift-i/cmd-shift-i) , then in the paths pallet create a path ("Make work path", iirc) for that selection .
