Course not. The Moscow metro was initially built as a huge bunker and in fact it really is. Actually a whole country is a network of bunkers, underground facilities food/fuel/weapon/clothes storages etc. For example, when I was in army, I ate a meat which was stored in one of those places since 1940. It wasn't as tasty like a fresh Angus steak, but I didn't die after it and had no diarrhea as well :) So be sure, even in case of total destruction, millions will survive.

The bombs tested on the surface back then were not anything close to the same weapons used today. All modern weapons were tested underground and yes the ground in those places including the ground water is very toxic.
